Research on effect and possible mechanisms of cutting formula of pantao pills on cognitive impairment in spontaneously hypertensive rats

Research on effect and possible mechanisms of cutting formula of pantao pills on cognitive impairment in spontaneously hypertensive rats

Abstract

Objective: To observe the effects of the cutting formula of Pantao Pills on the cognitive function of spontaneously hypertensive rats (SHRs), and the possible mechanism. Methods: Wistar kyoto (WKY) rats were used as control group. SHR were randomly divided into model group, irbesartan+donepezil group, donepezil group, low-dose group of the cutting formula of Pantao Pills 3.07 g/(kg·d) and high-dose group of the cutting formula of Pantao Pills 6.14 g/(kg·d). Twelve rats in each group, half male and half female, were given orally once a day for 11 weeks. After 11 weeks of administration, cognitive memory function was examined by Morris water maze; morphological changes of brain tissue were observed by hematoxylin-eosin (HE) staining; serum renin (REN) levels were measured by ELISA, and aldosterone (ALD) and angiotensin Ⅱ (Ang II) levels were measured by radioimmunoassay. Western Blot method was used to analyze phosphoinositide 3-kinase (PI3K), phosp-horylated-PI3K (p-PI3K), protein kinase B (AKT), p-AKT, extracellular signal-regulated kinase 1/2 (ERK1/2), p-ERK1/2, p38 mitogen-activated protein kinase (p38 MAPK), p-p38 MAPK protein expression level in brain. Results: Compared with the control group, the swimming distance of SHR model group was significantly longer; the cortex and hippocampus of SHR model rats were loose, the density of neurons was decreased, and there were more vacuoles and cell fragments, the levels of PI3K, p-AKT and AKT protein in brain tissue of SHR model group were significantly lower, while the levels of p-p38 MAPK protein were significantly higher; compared with SHR model group, the swimming distance of SHRs in the water maze positioning and navigation test was significantly shortened in the high dose group (P=0.01) and the low dose group (P=0.003) of the cutting formula of Pantao Pills, the high dose group and the low dose group of the cutting formula of Pantao Pills significantly reduced neuronal damage and nuclear cohesion in cortex and hippocampus, decreased the pathological changes of hippocampus and cortical neurons; the high dose group of the cutting formula of Pantao Pills can up-regulate the level of brain PI3K (7^=0.021), p-AKT (P=0.000), AKT (7^=0.001) protein, down-regulate the level of brain p-p38 MAPK (P=0.001), p38 MAPK (P=0.008) protein. Conclusion: This suggests that the cutting formula of Pantao Pills can improve the cognitive impairment of SHRs and protect neurons. The mechanism may be related to the regulation of PI3K/AKT, p-AKT, AKT, p-p38 MAPK and other related proteins in brain PI3K/AKT and p38 MAPK signaling pathways of SHRs.

Tau is the major microtubule associated protein (MAP) of a mature neuron. The other two neuronal MAPs are MAP1 and MAP2. An established function of MAPs is their interaction with tubulin and promotion of its assembly into microtubules and stabilization of the microtubule network. The microtubule assembly promoting activity of tau, a phosphoprotein, is regulated by its degree of phosphorylation. Normal adult human brain tau contains 2-3 moles phosphate/mole of tau protein. Hyperphosphorylation of tau depresses this biological activity of tau. In Alzheimer disease (AD) brain tau is ~three to four-fold more hyperphosphorylated than the normal adult brain tau and in this hyperphosphorylated state it is polymerized into paired helical filaments ([PHF) admixed with straight filaments (SF) forming neurofibrillary tangles. Tau is transiently hyperphosphorylated during development and during anesthesia and hypothermia but not to the same state as in AD brain. The abnormally hyperphosphorylated tau in AD brain is distinguished from transiently hyperphosphorylated tau by its ability (1) to sequester normal tau, MAP1 and MAP2 and disrupt microtubules, and (2) to self-assemble into PHF/SF. The cytosolic abnormally hyperphosphorylated tau, because of oligomerization, unlike normal tau, is sedimentable and on self-assembly into PHF/SF, loses its ability to sequester normal MAPs. Some of the tau in AD brain is truncated which also promotes its self-assembly. Tau mutations found in frontotemporal dementia apparently promote its abnormal hyperphosphorylation. Thus, the AD abnormally hyperphosphorylated tau (1) is distinguishable from both normal and transiently hyperphosphorylated taus, and (2) is inhibitory when in a cytosolic/oligomeric state but not when it is self-assembled into PHF/SF. Inhibition of abnormal hyperphosphorylation of tau offers a promising therapeutic target for AD and related tauopathies.

Human vascular smooth muscle cell proliferation and migration contribute to vascular remodeling in pulmonary hypertension and atherosclerosis. The precise mechanisms that regulate structural remodeling of the vessel wall remain unknown. This study tests the hypothesis that phosphatidylinositol 3-kinase (PI3K) activation is both necessary and sufficient to mediate human pulmonary vascular smooth muscle (PVSM) cell proliferation and migration. Microinjection of human PVSM cells with a dominant-negative class IA PI3K inhibited platelet-derived growth factor (PDGF)-induced DNA synthesis by 65% (P < 0.001; chi(2) analysis) compared with cells microinjected with control plasmid, whereas microinjection of cells with a constitutively active class IA PI3K (p110*-CA) was sufficient to induce DNA synthesis (mitotic index of p110*-CA-microinjected cells was 15% vs. 3% in control cells; P < 0.01). Transfection of PVSM cells with p110*-CA was also sufficient to promote human PVSM cell migration. In parallel experiments, stimulation of human PVSM cells with PDGF induced PI3K-dependent activation of Akt, p70 S6 kinase, and ribosomal protein S6 but not mitogen-activated protein kinase. PDGF-induced proliferation and migration was inhibited by LY-294002. These results demonstrate that PI3K signaling is both necessary and sufficient to mediate human PVSM cell proliferation and migration and suggest that the activation of PI3K may play an important role in vascular remodeling.
