Study on the inhibitory effect of Xiao-Xu-Ming decoction on ferritinophagy in Alzheimer's disease in vitro cell model

Abstract

Objective: To observe the effects of Xiao-Xu-Ming Decoction (XXMD) on the expression of nuclear receptor coactivator 4 (NCOA4) and ferritin heavy chain (FTH1) in glutamate (Glu)-induced neurotoxic injury in mouse hippocampal neuronal cells (HT22), and to explore the mechanism of traditional Chinese medicine in treating Alzheimer's disease (AD). Methods: HT22 cells in the logarithmic growth phase were divided into a control group, a Glu model group, and low- and high-dose XXMD groups. An in vitro AD cell model was established by inducing HT22 cell damage with 20 mmol/L Glu. The cells in the XXMD groups were treated with different concentrations (500 and 1 000 μg/mL) of XXMD. The effects of XXMD on the proliferation and survival rate were detected using the methyl thiazolyl tetrazolium (MTT) assay. Autophagy flux was assessed using mRFP-GFP-LC3 adenovirus transfection. The associations between NCOA4/LC3 and NCOA4/FTH1 expression were examined by immunofluorescence double staining. The protein expression levels of LC3BⅡ/Ⅰ, NCOA4, and FTH1 were measured using Western blot analysis. Results: XXMD alleviated Glu-induced damage to HT22 cells in a dose-dependent (P < 0.01). Compared with the control group, the Glu group showed increased autophagy flux, elevated protein expression of NCOA4 and LC3BⅡ/Ⅰ (P < 0.01), and decreased protein expression of FTH1 (P < 0.01). Compared with the Glu group, the low- and high-dose XXMD groups exhibited reduced autophagy flux, decreased protein expression of NCOA4 and LC3BⅡ/Ⅰ (P < 0.01), and upregulated FTH1 protein expression (P < 0.01). Conclusions: XXMD significantly improves the survival rate of Glu-induced HT22 cells and inhibits abnormal autophagy flux. The mechanism may involve the downregulation of NCOA4 protein expression and upregulation of FTH1 protein expression.

Alzheimer's disease (AD) has become a worldwide disease that is harmful to human health and brings a heavy economic burden to healthcare system. Xiao-Xu-Ming Decoction (XXMD) has been widely used to treat stroke and other neurological diseases for more than 1000 years in China. However, the synergistic mechanism of the constituents in XXMD for the potential treatment of AD is still unclear. Therefore, the present study aimed to predict the potential targets and uncover the material basis of XXMD for the potential treatment of AD. A network pharmacology-based method, which combined data collection, drug-likeness filtering and absorption, distribution, metabolism, excretion and toxicity (ADME/T) properties filtering, target prediction and network analysis, was used to decipher the effect and potential targets of XXMD for the treatment of AD. Then, the acetylcholinesterase (AChE) inhibitory assay was used to screen the potential active constituents in XXMD for the treatment of AD, and the molecular docking was furtherly used to identify the binding ability of active constituents with AD-related target of AChE. Finally, three in vitro cell models were applied to evaluate the neuroprotective effects of potential lead compounds in XXMD. Through the China Natural Products Database, Traditional Chinese Medicine Systems Pharmacology (TCMSP) Database, Traditional Chinese Medicine (TCM)-Database @Taiwan and literature, a total of 1481 compounds in XXMD were finally collected. After ADME/T properties filtering, 908 compounds were used for the further study. Based on the prediction data, the constituents in XXMD formula could interact with 41 AD-related targets. Among them, cyclooxygenase-2 (COX-2), estrogen receptor α (ERα) and AChE were the major targets. The constituents in XXMD were found to have the potential to treat AD through multiple AD-related targets. 62 constituents in it were found to interact with more than or equal to 10 AD-related targets. The prediction results were further validated by in vitro biology experiment, resulting in several potential anti-AD multitarget-directed ligands (MTDLs), including two AChE inhibitors with the IC50 values ranging from 4.83 to 10.22 μM. Moreover, fanchinoline was furtherly found to prevent SH-SY5Y cells from the cytotoxicities induced by sodium nitroprusside, sodium dithionate and potassium chloride. In conclusion, XXMD was found to have the potential to treat AD by targeting multiple AD-related targets and canonical pathways. Fangchinoline and dauricine might be the potential lead compounds in XXMD for the treatment of AD.

目的 检测HT22细胞系可否作为研究神经元铁死亡的良好细胞模型,在细胞水平上为研究神经元铁死亡的机制提供基础。方法 用经典铁死亡诱导剂Erastin和RSL3处理HT22细胞系,显微镜下观察细胞存活情况;使用CCK-8法检测细胞存活率;使用丙二醛法或菲罗嗪法分别检测细胞内脂质活性氧(reactive oxygen species,ROS)、Fe<sup>2+</sup>浓度;实时定量聚合酶链反应(quantitative real time polymerase chain reaction,qRT-PCR)法检测细胞铁死亡关键基因SLC7A11(solute carrier family 7 member 11)、PTGS2(prostaglandin-endoperoxide synthase 2)的mRNA水平。结果 Erastin或RSL3处理HT22后,显微镜下发现细胞发生明显死亡,加入铁死亡抑制剂Ferrostatin-1后,细胞死亡显著减少。Erastin或RSL3诱导后HT22细胞内的Fe<sup>2+</sup>浓度无明显变化(P>0.05),脂质过氧化水平明显增加(PSLC7A11、PTGS2的mRNA表达水平增加(P结论 HT22细胞系是一种铁死亡敏感细胞系,Erastin或RSL3诱导HT22铁死亡,HT22细胞系可用于研究神经元中铁死亡的发生及胞内脂质过氧化物蓄积机制。

Alzheimer’s disease (AD) is a progressive neurodegenerative disease characterized by memory loss and multiple cognitive impairments. Several decades of intense research have revealed that multiple cellular changes are implicated in the development and progression of AD, including mitochondrial damage, synaptic dysfunction, amyloid beta (Aβ) formation and accumulation, hyperphosphorylated tau (P-Tau) formation and accumulation, deregulated microRNAs, synaptic damage, and neuronal loss in patients with AD. Among these, mitochondrial dysfunction and synaptic damage are early events in the disease process. Recent research also revealed that Aβ and P-Tau-induced defective autophagy and mitophagy are prominent events in AD pathogenesis. Age-dependent increased levels of Aβ and P-Tau reduced levels of several autophagy and mitophagy proteins. In addition, abnormal interactions between (1) Aβ and mitochondrial fission protein Drp1; (2) P-Tau and Drp1; and (3) Aβ and PINK1/parkin lead to an inability to clear damaged mitochondria and other cellular debris from neurons. These events occur selectively in affected AD neurons. The purpose of our article is to highlight recent developments of a Aβ and P-Tau-induced defective autophagy and mitophagy in AD. This article also summarizes several aspects of mitochondrial dysfunction, including abnormal mitochondrial dynamics (increased fission and reduced fusion), defective mitochondrial biogenesis, reduced ATP, increased free radicals and lipid peroxidation, and decreased cytochrome c oxidase (COX) activity and calcium dyshomeostasis in AD pathogenesis. Our article also discusses how reduced levels of Drp1, Aβ, and P-Tau can enhance the clearance of damaged mitochondria and other cellular debris by autophagy and mitophagy mechanisms.

Increased levels of total tau (t-tau) and hyperphosphorylated tau (p-tau) proteins in the cerebrospinal fluid of Alzheimer's disease (AD) patients are well documented and strongly correlate with AD pathology. Recent studies have further shown that human tau can be released into the extracellular space and transferred to nascent neurons. However, because the tau protein has no signal peptide identity, the mechanisms underlying its secretion remain poorly understood. In the present study, we confirmed that tau protein secretion was promoted by autophagy inducers and downregulated by beclin1 knockdown or autophagy inhibitors derived from human wild type tau (wt-tau)-overexpressing SH-SY5Y cells. Moreover, both t-tau and p-tau secretion were increased by autophagy activation. Furthermore, we identified that six isoforms of tau protein are secreted in an autophagy-dependent manner. These results indicate that both normal and pathological tau are secreted via an autophagy-mediated secretory pathway in neurons. Understanding this new pathway for tau secretion may provide critical future insights into tau pathologies, such as AD.

A growing body of evidence suggests that autophagy inhibition enhances the effectiveness of chemotherapy, especially in difficult-to-treat cancers. Existing autophagy inhibitors are primarily lysosomotropic agents. More specific autophagy inhibitors are highly sought-after. The microtubule-associated protein 1A/1B light chain 3B protein, LC3B, is an adapter protein that mediates key protein-protein interactions at several points in autophagy pathways. In this work, we used a known peptide ligand as a starting point to develop improved LC3B inhibitors. We obtained structure-activity relationships that quantify the binding contributions of peptide termini, individual charged residues, and hydrophobic interactions. Based on these data, we used artificial amino acids and diversity-oriented stapling to improve affinity and resistance to biological degradation, while maintaining or improving LC3B affinity and selectivity. These peptides represent the highest-affinity LC3B-selective ligands reported to date, and they will be useful tools for further elucidation of LC3B's role in autophagy and in cancer.© 2020 Wiley-VCH Verlag GmbH & Co. Macroautophagy is a vacuolar degradation pathway that terminates in the lysosomal compartment. Macroautophagy is a multistep process involving: (1) signaling events that occur upstream of the molecular machinery of autophagy; (2) molecular machinery involved in the formation of the autophagosome, the initial multimembrane-bound compartment formed in the autophagic pathway; and (3) maturation of autophagosomes, which acquire acidic and degradative capacities. In this chapter we summarize what is known about the regulation of the different steps involved in autophagy, and we also discuss how macroautophagy can be manipulated using drugs or genetic approaches that affect macroautophagy signaling, and the subsequent formation and maturation of the autophagosomes. Modulating autophagy offers a promising new therapeutic approach to human diseases that involve macroautophagy.

In the past two decades, substantial effort has been made to identify the roles of autophagy in tumor suppression and promotion, neurodegenerative disorders, and other pathophysiologies. This review summarizes the current advances and discusses the unanswered questions in understanding the involvement of autophagy in pathogenic mechanisms of disease, primarily focusing on cancer and neurodegenerative diseases.
