Changes of Soil Biochemical Characteristics in Microbial Electrochemical Remediation of Contaminated Soil

Abstract

To explore the role of soil enzymes and pH in the degradation of tetracycline in Microbial Electrochemical Systems (MES), four types of soils including black soil, moisture soil, yellow brown soil and red soil were collected for study. The results showed that after adding tetracycline (TN treatment, 10 mg/kg) to black soil, moisture soil and red soil, the values of pH increased by 0.08, 0.15 and 1.09 units respectively compared with the control treatment (CK, without tetracycline addition). However, in MES treatment (TC, tetracycline concentration 10 mg/kg), the values of pH in the four types of soil all showed downward trend. Although the soil dehydrogenase, urease and polyphenol oxidase activities exhibited different variation trends in tetracycline contaminated soil, their activities in MES increased by 62.4%-72.1%, 11.3%-80.9% (except red soil) and 12.6%-42.6% compared with TN treatment, respectively. Meanwhile, the degradation rate of tetracycline in TC treatment was respectively increased by 14% (black soil), 26% (moisture soil), 26% (yellow brown soil) and 17% (red soil) compared with TN treatment. Correlation analysis indicated that dehydrogenase activity showed significant positive correlation with tetracycline degradation rate, while pH showed significant negative correlation with degradation rate, indicating that dehydrogenase might participate in the process of tetracycline degradation, and acidic conditions may be more conducive to degradation. This experiment revealed the degradation mechanism of tetracycline by MES based on soil biochemical characteristics, which could provide important support for soil pollution remediation.

Antibiotic-resistant pathogens are profoundly important to human health, but the environmental reservoirs of resistance determinants are poorly understood. The origins of antibiotic resistance in the environment is relevant to human health because of the increasing importance of zoonotic diseases as well as the need for predicting emerging resistant pathogens. This Review explores the presence and spread of antibiotic resistance in non-agricultural, non-clinical environments and demonstrates the need for more intensive investigation on this subject.

Antibiotics in soil environment are regarded as emerging pollutants and have introduced increasing risks to soil ecosystem and human health in rapid urbanization areas. Identifying the occurrence and spatial variability of antibiotics in soils is an urgent issue in sustaining soil security. In this study, antibiotics in soils were investigated and analyzed in Beijing-Tianjin-Hebei urban agglomeration. The occurrence, spatial distribution, and related affecting factors of antibiotics in soils were identified and ecological risks of antibiotics in soil environment were assessed. Results showed that (1) The mean concentration of soil antibiotics in Beijing-Tianjin-Hebei urban agglomeration was 21.79 µg/kg. Land use substantially affected the occurrence and concentration of antibiotics in soils. Concentrations of antibiotics in cropland and orchard soils were 2-3 times higher than the other land use types. (2) The concentrations of antibiotics in soils in Beijing-Tianjin-Hebei urban agglomeration presented a spatial pattern of high values in southeast, and low values in northwest. Spatial variability of antibiotics in soils was closely related to the application of organic fertilizer and wastewater irrigation as well as topographical features. Furthermore, soil properties and land management policy had substantial influences on soil antibiotics, and soil heavy metals may aggravate the accumulation of antibiotics in soils. (3) Ecological risks assessment of antibiotics in soils demonstrated that erythromycin (ERY), sulfamethoxazole (SMX), and doxycycline (DOX) may introduce high risks to soil ecosystem health, and more attention should be paid to the areas with intensive human activities that had potential high risk to soil ecosystem health. This study suggests that scientific land and soil management should be considered to prevent soil antibiotic pollution and sustain soil security in urban agglomeration.Copyright © 2022. Waste biomass is a cheap and relatively abundant source of electrons for microbes capable of producing electrical current outside the cell. Rapidly developing microbial electrochemical technologies, such as microbial fuel cells, are part of a diverse platform of future sustainable energy and chemical production technologies. We review the key advances that will enable the use of exoelectrogenic microorganisms to generate biofuels, hydrogen gas, methane, and other valuable inorganic and organic chemicals. Moreover, we examine the key challenges for implementing these systems and compare them to similar renewable energy technologies. Although commercial development is already underway in several different applications, ranging from wastewater treatment to industrial chemical production, further research is needed regarding efficiency, scalability, system lifetimes, and reliability.

Antibiotics and antibiotic resistance genes (ARGs) are two pollutants in soil, especially ARGs as one of the top three threats to human health. The performance of soil microbial fuel cells (MFCs) fuelled with antibiotics was investigated.In this study, soil MFCs spiked with tetracycline exhibited optimal bioelectricity generation, which was 25% and 733% higher than those of MFCs spiked with sulfadiazine and control, respectively. Compared with the non-electrode treatment, not only did functional micro-organisms change in open- and closed-circuit treatments, but also the microbial affinities, respectively, increased by 50% and 340% to adapt to higher removal of antibiotics. For the open-circuit treatment, the ineffective interspecific relation of micro-organisms was reduced to assist the removal efficiency of antibiotics by 7-27%. For the closed-circuit treatment, an intensive metabolic network capable of bioelectricity generation, degradation and nitrogen transformation was established, which led to 10-35% higher removal of antibiotics. Importantly, the abundances of ARGs and mobile genetic element (MGE) genes decreased after the introduction of electrodes; especially in the closed-circuit treatment, the highest reduction of 47% and 53% was observed, respectively.Soil MFCs possess advantages for the elimination of antibiotics and ARGs with sevenfold to eightfold higher electricity generation than that of the control treatment. Compared with sulphonamides, the enhancement removal of tetracycline is higher, while both potential ARG propagation risk is reduced in soil MFCs. This study firstly synchronously reveals the relationships among bacteria, fungi and archaea and with ARGs and MGE genes in soil bioelectrochemical systems.

Exposure to tetracycline in soil causes microbial mutations. Soil microbial fuel cells (MFCs) can promote the degradation efficiency of contaminants while generating bioelectricity under anaerobic conditions. MFC performance varies amongst different types of soils due to distinctive soil properties. This study assesses the performance of soil MFCs filled with four typical Chinese soils and explores key factors regulating bioelectricity generation and tetracycline degradation. Except for the MFCs filled with black soil, tetracycline degradation rates improved in soil MFCs, particularly in those filled with Chao soil, which enhanced the degradation rate by 39% relative to the corresponding control. Additionally, soil MFCs filled with Chao soil exhibited the highest charge output of 1347 ± 357C, which was 100-499% higher than that of MFCs with other soils. According to redundancy analysis, soil particle size, pH, conductivity and dissolved organic carbon content showed positive association with tetracycline degradation and charge output, while the adsorption of tetracycline had a negative association with degradation rate. Thus, the adsorption of tetracycline restricted its removal efficiency in soil MFCs, and high soil conductivity and large particle size promoted electron transfer, enhancing biocurrent intensity, which increased tetracycline degradation efficiency.Copyright © 2019 Elsevier B.V. Soil contaminated by aged petroleum hydrocarbons is faced with scarcity of electron acceptors, low activity of functional microbes and inefficient electron transfer, which hinder the bioremediation application. The soil microbial fuel cell (MFC) simultaneously solves these problems with bioelectricity production. In this study, five types of surfactants were introduced to enhance the bioavailability of aged petroleum hydrocarbon in soils. The ampholytic surfactant (lecithos) was optimal due to the highest bioelectricity generation (0.321Cdg) and promoted hydrocarbon degradation (328%), while the nonionic (glyceryl monostearate) and cationic (cetyltrimethylammonium bromide) surfactants were inefficient. The surfactants induced a special microbial enrichment affiliated with Proteobacteria, Firmicutes, Bacteroidetes, Actinobacteria, Chloroflexi, Planctomycetes and Acidobacteria (93%-99% of total) in soil MFCs. The anionic surfactant (sodium dodecyl sulfate) exhibited the strongest selectivity, and α-proteobacteria and γ-proteobacteria abundances decreased while Clostridia increased, much like the result obtained with the biosurfactant β-cyclodextrin. Furthermore, Bacillus abundance was increased in connected soil MFCs, except addition of lecithos in which Clostridium increased to 14.88% from 3.61% in the control. The high correlations among Bacillus, Phenylobacterium, Solibacillus (0.9162-0.9577) and among Alcaligenes, Dysgonomonas, Sedimentibacter (0.9538-0.9966) indicated a metabolic network of microorganisms in the soil bioelectrochemical remediation system.Copyright © 2017 Elsevier B.V.
