Identification of Tomato Spotted Wilt Orthotospovirus on Tomato and Pepper in Shaanxi

Abstract

In order to definite the species of virus, which cause tomato leaf samples chlorosis necrosis, fruit necrosis, and concentric spots, we collected the susceptible tomato samples, and pepper leaf samples exhibiting symptoms of brown wilting and necrosis, fruit deformity and shriveling. RT-PCR detection was performed utilizing specific primers targeting the tomato spotted wilt virus (TSWV) as well as N gene primers. The results indicated that occurrence of TSWV infection in both tomato and pepper samples. Phylogenetic analysis based on the N gene sequence demonstrated a close relationship between TSWV isolates obtained from tomatoes and peppers in Shaanxi with those found in Australian peppers, Japanese tomatoes, as well as Beijing Verbenas within China. During the virus inoculation experiment, tomato and pepper samples both displayed characteristic symptoms including chlorosis, necrotic foliage, and shriveled fruits; RT-PCR successfully detected the presence of TSWV in these plants. This study confirms that tomatoes and peppers in Shaanxi have been affected by TSWV infection and provides a theoretical basis for its prevention and control.

【目的】 对我国茄科、葫芦科、豆科和十字花科蔬菜主要病毒病开展调查、诊断,明确当前我国主要蔬菜作物病毒病的病原物、优势病毒及其分布,并对其中一些重要病毒引起的病毒病在我国的发生危害趋势进行分析。【方法】 2013—2017年,对我国大陆31个省(市、自治区)开展茄科、葫芦科、豆科和十字花科等蔬菜作物主要病毒病发生情况的普查,利用血清学和分子生物学相结合的方法对主要蔬菜病毒病的病原进行鉴定。【结果】 我国大陆31个省(市、自治区)共采集茄科、葫芦科、豆科和十字花科蔬菜作物疑似病毒病样品41 653份,共检出病毒63种,其中茄科蔬菜检出病毒达40种,葫芦科蔬菜检出病毒26种,豆科蔬菜检出病毒19种,十字花科蔬菜检出病毒14种。茄科的辣椒检出病毒33种,番茄检出病毒25种;葫芦科的南瓜检出病毒22种,黄瓜检出病毒19种;豆科的豇豆检出病毒14种,菜豆检出病毒12种;十字花科的萝卜检出12种病毒,大白菜检出7种病毒。蔬菜作物中普遍存在多种病毒复合侵染现象,已发现2—6种病毒复合侵染,以2种病毒复合侵染类型居多,其中辣椒、番茄和茄子上存在6种病毒复合侵染。本研究首次在国内发现番茄斑驳花叶病毒(Tomato mottle mosaic virus,ToMMV)、西葫芦虎纹花叶病毒(Zucchini tigre mosaic virus,ZTMV)、辣椒脉黄化病毒(Pepper vein yellows virus,PeVYV)、辣椒隐潜病毒1号(Pepper cryptic virus 1,PCV-1)和辣椒隐潜病毒2号(Pepper cryptic virus 2,PCV-2)的侵染;首次发现ToMMV、甜瓜蚜传黄化病毒(Melon aphid-borne yellows virus,MABYV)、南瓜蚜传黄化病毒(Cucurbit aphid-borne yellows virus,CABYV)对辣椒的侵染,首次发现ZTMV对黄瓜、烟草轻绿花叶病毒(Tobacco mild green mosaic virus,TMGMV)对南瓜的侵染,首次发现番茄斑萎病毒(Tomato spotted wilt tospovirus,TSWV)可以侵染芹菜、曼陀罗、豇豆、豌豆、党参、大丽花、旱金莲、刺天茄等,首次发现红辣椒脉斑驳病毒(Chilli veinal mottle virus,ChiVMV)可以侵染红茄,另外还首次发现辣椒和番茄为烟草丛顶病毒(Tobacco bushy top virus,TBTV)的新自然寄主。【结论】 黄瓜花叶病毒(Cucumber mosaic virus,CMV)、烟草花叶病毒(Tobacco mosaic virus,TMV)、芜菁花叶病毒(Turnip mosaic virus,TuMV)和蚕豆萎蔫病毒2号(Broad bean wilt virus 2,BBWV2)为当前危害我国蔬菜作物的优势病毒,在全国范围内广泛分布且发生严重,特别是CMV发生最为普遍,在31个省(市、自治区)均有分布,且均为这些地区的优势病毒。茄科蔬菜的优势病毒为CMV和TMV,葫芦科蔬菜的优势病毒依次为CMV、小西葫芦黄花叶病毒(Zucchini yellow mosaic virus,ZYMV)、黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)和TMV,豆科蔬菜的优势病毒为CMV和BBWV2,十字花科蔬菜的优势病毒依次为TuMV、CMV和TMV。番茄褪绿病毒(Tomato chlorosis virus,ToCV)、TSWV、CGMMV和ToMMV等病毒在部分省区发生严重,扩展蔓延速度极快,具有在全国范围内流行的风险。

利用番茄斑萎病毒(tomato spotted wilt orthotospovirus,TSWV)通用引物及自行设计合成引物对采集自内蒙古自治区赤峰市部分地区的番茄病毒病样品进行了RT-PCR检测及序列测定;进一步利用特异性引物对包含N基因在内核苷酸序列进行扩增、克隆和测序,并对包含N基因的914 nt序列进行系统进化分析。结果表明,在采集的16个表现TSWV症状和2个未表现典型症状的果实样品中均检测到TSWV;TSWV内蒙古番茄分离物与中国云南、江苏等地的分离物亲缘关系最近。用表现TSWV症状的番茄果实汁液摩擦接种辣椒叶片,出现叶片褪绿、植株矮小等症状,并经RT-PCR分析检测到TSWV。本研究证实在内蒙古地区有TSWV在番茄上发生危害。

Tomato spotted wilt virus (TSWV) is transmitted by Frankliniella occidentalis (Pergande) in a persistent-propagative manner. We previously observed significant results in terms of feeding behavior of spinosad-susceptible (Ivf03) and -resistant (Spin-R) strains of F. occidentalis using electrical penetration graph. TSWV transmission by the two strains was compared in the present study. The results showed that the titer of TSWV-N RNA (a part of S RNA of TSWV and encoding the nucleocapsid protein) in Ivf03 and Spin-R strains was not significantly different after a 48-h inoculation access period. The TSWV transmission rate did not significantly differ between the two strains and was 51.0% for Ivf03 and 44.4% for Spin-R. The virus transmission rate was significantly higher for males than females of both strains. The virus transmission rate for males and females of Ivf03 was 68.1 and 33.8%, respectively; however, in case of Spin-R, it was 60 and 28.8% for males and females, respectively. Additionally, number of probes and duration of probes were generally greater for viruliferous females of Ivf03 than for viruliferous females of Spin-R but the total number and duration of noningestion probes did not significantly differ between males of the two strains. The latter finding behavior may help explain the similar transmission rates for the susceptible and resistant strains. © The Authors 2015. A method is presented for the rapid isolation of high molecular weight plant DNA (50,000 base pairs or more in length) which is free of contaminants which interfere with complete digestion by restriction endonucleases. The procedure yields total cellular DNA (i.e. nuclear, chloroplast, and mitochondrial DNA). The technique is ideal for the rapid isolation of small amounts of DNA from many different species and is also useful for large scale isolations.

为了检测和鉴定我国不同地区番茄上的病毒种类,2015-2018年,在不同蔬菜种植区采集431份番茄疑似感病样品。利用已报道侵染番茄的12种主要病毒的特异性引物对样品进行RT-PCR分子检测与鉴定,结果表明:TYLCV在番茄上普遍存在,检出率为65.20%;而ToCV、TMV、CMV、ToMV、TSWV和PVY侵染番茄相对较少,检出率分别为21.11%,20.19%,17.63%,15.31%,14.15%,11.37%;未检测到TICV、PVX、TYLCVNB和BBWV等病毒。通过对番茄病毒复合侵染现象进行分析发现,病毒复合侵染率达35.73%,其中2种病毒复合侵染现象最多,占复合侵染总比的52.60%;3种病毒复合侵染率占复合侵染总比27.92%;4种及5种病毒复合侵染率略低,分别占复合侵染总比0.65%和18.83%。通过对我国20个地区的番茄病毒进行检测,结果表明,在番茄上检出病毒种类最多的为北京地区,检测到7种病毒,其次是山东地区,检测到5种病毒;河南、甘肃及宁夏地区检测到4种病毒;西藏、河北、浙江、陕西、海南地区检测到3种病毒;四川、天津、黑龙江、山西、内蒙古等检测到2种病毒;其他地区只检测到单一番茄病毒种类。同时根据TYLCV和TSWV部分基因序列构建系统发育树,结果表明,TYLCV北京分离物(MK336782)与中国湖南分离物的亲缘关系最近,TSWV河南分离物(MK318839)与山东分离物的亲缘关系最近。

在对青海辣椒上的病毒病进行调查时发现疑似番茄斑萎病毒(Tomato spotted wilt tospovirus,TSWV)病样,表现为环斑、坏死等症状。为明确其感染的病毒种类,对其进行了检测和鉴定。对Tospovirus的通用引物进行检测时发现采集的49份样品中有14份样品扩增到了目的条带,序列测定结果显示其与TSWV的同源性最高;同时利用TSWV特异性引物对阳性样品进行检测,结果全部为阳性;基于克隆的NSs基因序列聚类分析结果显示其属于TSWV的1个分离物,与中国云南、黑龙江等地分离物相对近缘,而与国外其他分离物相对远缘。在分子鉴定的基础上,利用TSWV的抗体通过Western blot进一步对样品进行了检测,结果也证明采集样品中存在TSWV的感染。这些结果表明青海辣椒上存在TSWV的感染。

为明确重庆地区辣椒上番茄斑萎病毒(Tomato spotted wilt virus,TSWV)的发生情况及其外壳蛋白基因(cp)序列变异特征,采用斑点免疫杂交法(Dot-ELISA)和反转录PCR(RT-PCR)技术对采自重庆的298份辣椒病毒病样品进行了检测和分子鉴定。Dot-ELISA检测结果表明,有40份辣椒样品检测到TSWV,检出率为13.42%。通过电子显微镜在TSWV检测呈阳性的样品中观察到具包膜的球状病毒粒体。设计扩增TSWV cp的引物,选取6个TSWV阳性样品进行RT-PCR扩增,均扩增到大小约750 bp的目的片段。序列分析表明,重庆辣椒分离物TSWV-CQ(KX611497)与已报道的22个TSWV分离物cp基因的核苷酸相似性为96.1% ~ 99.4%,与已报道的TSWV中国分离物亲缘关系较近,表现出明显的地理相关性。
