Study on Lignification Dynamics of Dendrocalamus brandisii Shoots Under Low Temperature Preservation After Harvesting

Abstract

In order to provide a theoretical basis for the preservation technology of Dendrocalamus brandisii shoots after picking, the alleviating effects of different low temperature preservation methods on the lignification process of D. brandisii shoots after harvesting were investigated. In this study, the bamboo shoots of D. brandisii from low hilly forest in Guangxi were used as research materials to analyze the changes of lignin, crude fiber content and key enzyme activity of lignin synthesis within 0-10 days after low temperature (4℃) vacuum storage and low temperature sealed storage. The results showed that the contents of lignin and crude fiber in bamboo shoots increased in different degrees within 1-10 d under the two cryopreservation methods. Vacuum treatment could effectively inhibit the synthesis of 43.21% lignin and 28.61% crude fiber. The total phenol content under vacuum treatment always maintained a continuous downward trend, while the total phenolic content decreased under sealing treatment until the 5th day and showed a certain degree of recovery. The phenylalanine ammonia lyase (PAL) activity of bamboo shoots under the two treatments showed a trend of decreasing first and then slowly increasing within 0-2 d, and the PAL activity under vacuum treatment was significantly lower than that under sealing treatment from the 3rd day (P<0.001). Principal component analysis and correlation analysis showed that lignin, crude fiber and PAL activity were positively correlated with storage days, and negatively correlated with total phenols. The results showed that total phenol, lignin, crude fiber content and PAL activity were the main response indexes in the lignification process of D. brandisii shoots after harvesting. The physical preservation method of 4℃ low temperature vacuum preservation can effectively delay the lignification process and the rapid decline of taste quality of bamboo shoots.

Plant biomass contains lignin that can be converted into high-value-added chemicals, fuels, and materials. The precise genetic manipulation of lignin content and composition in plant cells offers substantial environmental and economic benefits. However, the intricate regulatory mechanisms governing lignin formation challenge the development of crops with specific lignin profiles. Mathematical models and computational simulations have recently been employed to gain fundamental understanding of the metabolism of lignin and related phenolic compounds. This review article discusses the strategies used for modeling plant metabolic networks, focusing on the application of mathematical modeling for flux network analysis in monolignol biosynthesis. Post-translational modification of proteins exerts essential roles in many biological processes in plants. The function of these chemical modifications has been extensively characterized in many physiological processes, but how these modifications regulate lignin biosynthesis for wood formation remained largely unknown. Over the past decade, post-translational modification of several proteins has been associated with lignification. Phosphorylation, ubiquitination, glycosylation, and S-nitrosylation of transcription factors, monolignol enzymes, and peroxidases were shown to have primordial roles in the regulation of lignin biosynthesis. Pomegranate is a subtropical and tropical fruit of great importance from a health point of view. Despite increasing consumer awareness of the health benefits of pomegranate, consumption of the fruit is still limited owing to poor postharvest handling, storage recommendations, short shelf life and quality deterioration during transportation, storage and marketing. The occurrence of physiological disorders such as husk scald, splitting and chilling injury is another challenge reducing marketability and consumer acceptance. Recently, notable work on postharvest biology and technology has been done. Pomegranate is highly sensitive to low-oxygen (<5 kPa) atmospheres, chilling injury and decay. One of the major problems associated with pomegranate fruit is excessive weight loss, which may result in hardening of the husk and browning of the rind and arils. To reduce chilling injury incidence and to extend storability and marketing of pomegranates, good results were obtained with polyamine, heat, salicylic acid, methyl jasmonate or methyl salicylate treatments prior to cold storage. This article reviews the maturity indices, changes during maturation and ripening, postharvest physiology and technology of pomegranate fruit as well as the various postharvest treatments for maintaining fruit quality.© 2015 Society of Chemical Industry.

在4 ℃,相对湿度85%条件下,以感官评分初步评判绿竹笋的储藏期限,结合亮度,失重率,呼吸强度,总酸,纤维素含量,木质素含量,亚硝酸盐含量及微生物数量等指标评价保鲜效果。结果表明:无包装绿竹笋储藏期为12 d,普通真空包装绿竹笋的储藏期为16 d,纳米真空包装的绿竹笋可储藏20 d。储藏第20天时,Ag-SiO<sub>2</sub>-TiO<sub>2</sub>纳米包装绿竹笋的感官评分为75.25分,亮度值为86.85,总酸含量为0.54 g/kg,纤维素含量为2.96 mg/g,木质素含量为0.72 mg/g。在4 ℃条件下,纳米真空包装处理可以保证绿竹笋的良好感官品相和可食用性,延长绿竹笋的储藏期限。因此,Ag-SiO<sub>2</sub>-TiO<sub>2</sub>纳米材料真空包装处理是一种良好的绿竹笋贮藏保鲜方法。

Lignin is a polyphenolic polymer that strengthens and waterproofs the cell wall of specialized plant cell types. Lignification is part of the normal differentiation programme and functioning of specific cell types, but can also be triggered as a response to various biotic and abiotic stresses in cells that would not otherwise be lignifying.Cell wall lignification exhibits specific characteristics depending on the cell type being considered. These characteristics include the timing of lignification during cell differentiation, the palette of associated enzymes and substrates, the sub-cellular deposition sites, the monomeric composition and the cellular autonomy for lignin monomer production. This review provides an overview of the current understanding of lignin biosynthesis and polymerization at the cell biology level.The lignification process ranges from full autonomy to complete co-operation depending on the cell type. The different roles of lignin for the function of each specific plant cell type are clearly illustrated by the multiple phenotypic defects exhibited by knock-out mutants in lignin synthesis, which may explain why no general mechanism for lignification has yet been defined. The range of phenotypic effects observed include altered xylem sap transport, loss of mechanical support, reduced seed protection and dispersion, and/or increased pest and disease susceptibility.© The Author 2015.
