Potential Mechanism of Yeast Improving Production Performance of Laying Hens Based on Transcriptome Technology

Abstract

This study aims to elucidate the potential mechanisms of yeast supplementation in diets for laying hens to improve their production performance and solve the problem of unclear yeast working mechanism using transcriptomics and genomics technologies. Compared to the basal diet group, yeast supplementation in laying hen diets increased the Chao and Shannon indices of the cecal microbiota (P>0.05). At the phylum level, the relative abundance of Bacteroidetes and Firmicutes increased in the yeast-supplemented group (P>0.05). At the genus level, the relative abundance of Bacteroides, Lactobacillus, Parabacteroides distasonis, Prevotella, Ruminococcus, Faecalibacterium, and Phascolarctobacterium also increased (P>0.05). Additionally, the relative abundance of Rikenella was significantly decreased compared to the basal diet group (P≤0.05). Transcriptomic analysis revealed that 329 genes were differentially expressed in ileal mucosal tissues, with 184 genes upregulated and 145 genes down regulated. These changes together improved the absorption and utilization efficiency of nutrients in diets for laying hens. This study revealed the molecular mechanism of yeast in improving the diets utilization rate of laying hens by regulating the structure of cecal microbiota and the expression of key genes in ileum mucosa, which provided a theoretical basis for the application of yeast as a green and safe new antibiotic substitute.

A key aspect of bacterial survival is the ability to evolve while migrating across spatially varying environmental challenges. Laboratory experiments, however, often study evolution in well-mixed systems. Here, we introduce an experimental device, the microbial evolution and growth arena (MEGA)-plate, in which bacteria spread and evolved on a large antibiotic landscape (120 × 60 centimeters) that allowed visual observation of mutation and selection in a migrating bacterial front. While resistance increased consistently, multiple coexisting lineages diversified both phenotypically and genotypically. Analyzing mutants at and behind the propagating front, we found that evolution is not always led by the most resistant mutants; highly resistant mutants may be trapped behind more sensitive lineages. Exopolysaccharides (EPS) are extracellular macromolecules excreted as tightly bound capsule or loosely attached slime layer in microorganisms. They play most prominent role against desiccation, phagocytosis, cell recognition, phage attack, antibiotics or toxic compounds and osmotic stress. In the last few decades, natural polymers have gained much attention among scientific communities owing to their therapeutic potential. In particular the EPS retrieved from probiotic bacteria with varied carbohydrate compositions possess a plenty of beneficial properties. Different probiotic microbes have unique behavior in expressing their capability to display significant health promoting characteristics in the form of polysaccharides. In this new era of alternative medicines, these polysaccharides are considered as substitutes for synthetic drugs. The EPS finds applications in various fields like textiles, cosmetics, bioremediation, food and therapeutics. The present review is focused on sources, chemical composition, biosynthetic pathways of EPS and their biological potential. More attention has been given to the scientific investigations on antimicrobial, antitumor, anti-biofilm, antiviral, anti-inflammatory and immunomodulatory activities.Copyright © 2020 Elsevier B.V. Although the production of safe food for human consumption is the primary purpose for animal rearing, the environment and well-being of the animals must also be taken into consideration. Based on microbiological point of view, the production of healthy food from animals involves considering foodborne pathogens, on the one hand and on the other hand, the methods used to fight against germs during breeding. The conventional method to control or prevent bacterial infections in farming is the use antibiotics. However, the banning of these compounds as growth promoters caused many changes in animal breeding and their use has since been limited to the treatment and prevention of bacterial infections. In this function, their importance no longer needs to be demonstrated, but unfortunately, their excessive and abusive use have led to a double problem which can have harmful consequences on consumer health: Resistance to antibiotics and the presence of antibiotic residues in food. The use of probiotics appears to be a suitable alternative to overcome these problems because of their ability to modulate the immune system and intestinal microflora, and further considering their antagonistic role against certain pathogenic bacteria and their ability to play the role of growth factor (sometimes associated with prebiotics) when used as feed additives. This review aims to highlight some of the negative effects of the use of antibiotics in animal rearing as well as emphasize the current knowledge on the use of probiotics as a feed additive, their influence on animal production and their potential utility as an alternative to conventional antibiotics, particularly in poultry, pig, and fish farming.Copyright: © Arsène, et al.

This research investigated effects of dietary phytosterols supplementation on growth performance and cecal gut microflora in yellow-feather broilers. A total of 360 yellow-feather broilers (1-day-old) were randomly assigned to 3 treatment groups: control group (basal diet), antibiotic group (basal diet supplemented with 200 mg/kg oxytetracycline calcium and 250 mg/kg nosiheptide), and phytosterols groups (basal diet supplemented with 25 mg/kg phytosterols). Each treatment group had 6 replicates, and there were 20 broilers within each replicate. No treatment effects on average daily feed intake, average daily gain, and food conversion rate were observed. The antibiotic group had a lower liver index compared with control group and phytosterols group. Other visceral indexes including bursa of Fabricius, spleen, and heart were not different among the 3 treatment groups. In terms of alpha diversity, no treatment effects on Shannon and Simpson indexes were observed. Supplementation of phytosterols significantly decreased the Chao1 and Ace indexes, indicating lower community richness of the gut microflora. At phylum level, the phytosterols group had a higher abundance of Bacteroidetes compared with the control group. At genus level, no treatment effect was observed on the top 10 genera. Overall, supplementation of phytosterols at 25 mg/kg level did not affect the growth performance of yellow-feather broilers, and its effect on gut microflora was limited.Copyright © 2020. The study of the extreme limits of human lifespan may allow a better understanding of how human beings can escape, delay, or survive the most frequent age-related causes of morbidity, a peculiarity shown by long-living individuals. Longevity is a complex trait in which genetics, environment, and stochasticity concur to determine the chance to reach 100 or more years of age [1]. Because of its impact on human metabolism and immunology, the gut microbiome has been proposed as a possible determinant of healthy aging [2, 3]. Indeed, the preservation of host-microbes homeostasis can counteract inflammaging [4], intestinal permeability [5], and decline in bone and cognitive health [6, 7]. Aiming at deepening our knowledge on the relationship between the gut microbiota and a long-living host, we provide for the first time the phylogenetic microbiota analysis of semi-supercentenarians, i.e., 105-109 years old, in comparison to adults, elderly, and centenarians, thus reconstructing the longest available human microbiota trajectory along aging. We highlighted the presence of a core microbiota of highly occurring, symbiotic bacterial taxa (mostly belonging to the dominant Ruminococcaceae, Lachnospiraceae, and Bacteroidaceae families), with a cumulative abundance decreasing along with age. Aging is characterized by an increasing abundance of subdominant species, as well as a rearrangement in their co-occurrence network. These features are maintained in longevity and extreme longevity, but peculiarities emerged, especially in semi-supercentenarians, describing changes that, even accommodating opportunistic and allochthonous bacteria, might possibly support health maintenance during aging, such as an enrichment and/or higher prevalence of health-associated groups (e.g., Akkermansia, Bifidobacterium, and Christensenellaceae).Copyright © 2016 Elsevier Ltd. The incidence of colon cancer has increased considerably and the intestinal microbiota participate in the development of colon cancer. We showed that the L. casei Zhang or vitamin K2 (Menaquinone-7) intervention significantly alleviated intestinal tumor burden in mice. This was associated with increased serum adiponectin levels in both treatments. But osteocalcin level was only increased by L. casei Zhang. Furthermore, the anti-carcinogenic actions of L. casei Zhang were mediated by hepatic Chloride channel-3(CLCN3)/Nuclear Factor Kappa B(NF-κB) and intestinal Claudin15/Chloride intracellular channel 4(CLIC4)/Transforming Growth Factor Beta(TGF-β) signaling, while the vitamin K2 effect involved a hepatic Vitamin D Receptor(VDR)-phosphorylated AMPK signaling pathway. Fecal DNA sequencing by the Pacbio RSII method revealed there was significantly lower Helicobacter apodemus, Helicobacter mesocricetorum, Allobaculum stercoricanis and Adlercreutzia equolifaciens following both interventions compared to the model group. Moreover, different caecum acetic acid and butyric acid levels and enrichment of other specific microbes also determined the activity of the different regulatory pathways. Together these data show that L. casei Zhang and Vitamin K2 can suppress gut risk microbes and promote beneficial microbial metabolites to reduce colonic tumor development in mice.
