Panax notoginseng Saponins: Bibliometric Analysis and Visual Analysis Based on Knowledge Graph Analysis

Panax notoginseng Saponins: Bibliometric Analysis and Visual Analysis Based on Knowledge Graph Analysis

Abstract

The purpose of this study is to systematically analyze the research hotspots and evolutionary trends of Panax notoginseng saponins (PNS) from 2000 to 2024 by combining knowledge graph with bibliometric methods. A total of 769 Chinese-language literatures and 300 English-language literatures related to PNS published from 2000 to 2024 were retrieved from the Web of Science Core Collection and China National Knowledge Infrastructure (CNKI), and knowledge graphs were constructed and bibliometric analysis was performed using tools such as VOSviewer, CiteSpace and R programming language. The results showed that the annual number of publications had a steady overall growth from 2000 to 2024, with a relatively rapid increase in English-language literatures in recent years; core authors and institutions had formed a close cooperative network. High-frequency keywords focused on themes such as Panax notoginseng, saponin components, pharmacological effects and quality control. Cluster analysis revealed research modules including "Extraction, Purification and Analysis", "Mechanism of Action and Pharmacodynamics" and "Network Pharmacology". Burst keywords and time-zone maps showed that the research focus had evolved from basic components and applications in cerebrovascular diseases to mechanisms of apoptosis and oxidative stress, and then to emerging directions such as pharmacokinetics and autophagy. The knowledge graph revealed that the research on Panax notoginseng saponins had formed a research network with Chinese universities and research institutes as the core and multi-country collaboration. Beijing University of Chinese Medicine, Chinese Academy of Chinese Medical Sciences and other institutions had made outstanding contributions. The research focused on cardiovascular and cerebrovascular protection, anti-inflammatory, anti-oxidation, anti-apoptosis, autophagy regulation and so on. Accurate delivery, multi-target coordination, and intestinal flora regulation are key breakthrough areas in the future. This study comprehensively combs the research context and frontier of Panax notoginseng saponins, which can provide quantitative reference and direction guidance for subsequent basic research, clinical transformation and industrial development.

三七中发挥药效的主要成分为三七皂苷R1、人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1和人参皂苷Rd,用于贫血、冠心病、高血压、脑卒中后遗症等疾病的治疗,但其化学成分多且难分离。将氢化松香丙烯酸羟乙酯(HRHA)通过巯基-烯点击化学反应键合到烷基化硅胶表面,制备出一种新型的改性松香键合二氧化硅高效液相色谱固定相(SiO<sub>2</sub>@HRHA),用于三七总皂苷的分离。对色谱固定相进行一系列表征,表明SiO<sub>2</sub>@HRHA固定相具有球形度好和表面多孔的特点,比表面积为308.55 m<sup>2</sup>/g,平均孔径达到6.78 nm。将制备的固定相湿法装柱,色谱柱性能评价结果表明,SiO<sub>2</sub>@HRHA柱具备反相色谱行为、较好的流通性及重复性。从色谱柱的综合评价Tanaka测试可知,SiO<sub>2</sub>@HRHA柱具有较好的立体选择性及氢键容量。将其用于三七总皂苷的分离,SiO<sub>2</sub>@HRHA柱在一定程度上优于C18柱,而SiO<sub>2</sub>@HRHA柱的分离效果又优于实验室同期合成的色谱柱,对5种皂苷(R1、Rg1、Re、Rb1和Rd)的分离度依次为3.33、3.54、20.17和9.72,表明SiO<sub>2</sub>@HRHA高效液相色谱柱对三七总皂苷具有优异的分离效果,为从实际样品中分离纯化三七总皂苷提供了新思路。

Diabetes mellitus (DM) is a chronic metabolic disorder associated with elevated blood glucose levels due either to insufficient insulin production (type 1 DM) or to insulin resistance (type 2 DM). The incidence of DM around the world continues to rise dramatically with more than 400 million cases reported today. Among the most serious consequences of chronic DM are cardiovascular complications that can have deleterious effects. Although numerous treatment options are available, including both pharmacological and nonpharmacological, there is substantial emerging interest in the use of traditional medicines for the treatment of this condition and its complications. Among these is ginseng, a medicinal herb that belongs to the genus Panax and has been used for thousands of years as a medicinal agent especially in Asian cultures. There is emerging evidence from both animal and clinical studies that ginseng, ginseng constituents including ginsenosides, and ginseng-containing formulations can produce beneficial effects in terms of normalization of blood glucose levels and attenuation of cardiovascular complications through a multiplicity of mechanisms. Although more research is required, ginseng may offer a useful therapy for the treatment of diabetes as well as its complications.

Cerebral ischemia-reperfusion is a complicated pathological process. The injury and cascade reactions caused by cerebral ischemia and reperfusion are characterized by high mortality, high recurrence, and high disability. However, only a limited number of antithrombotic drugs, such as recombinant tissue plasminogen activator (r-TPA), aspirin, and heparin, are currently available for ischemic stroke, and its safety concerns is inevitable which associated with reperfusion injury and hemorrhage. Therefore, it is necessary to further explore and examine some potential neuroprotective agents with treatment for cerebral ischemia and reperfusion injury to reduce safety concerns caused by antithrombotic drugs in ischemic stroke. Ginseng Rg1 (G-Rg1) is a saponin composed of natural active ingredients and derived from the roots or stems of Panax notoginseng and ginseng in traditional Chinese medicine. Its pharmacological effects exert remarkable neurotrophic and neuroprotective effects in the central nervous system. To explore and summarize the protective effects and mechanisms of ginsenoside Rg1 against cerebral ischemia and reperfusion injury, we conducted this review, in which we searched the PubMed database to obtain and organize studies concerning the pharmacological effects and mechanisms of ginsenoside Rg1 against cerebral ischemia and reperfusion injury. This study provides a valuable reference and clues for the development of new agents to combat ischemic stroke. Our summarized review and analysis show that the pharmacological effects of and mechanisms underlying ginsenoside Rg1 activity against cerebral ischemia and reperfusion injury mainly involve 4 sets of mechanisms: anti-oxidant activity and associated apoptosis via the Akt, Nrf2/HO-1, PPARγ/HO-1, extracellular regulated protein kinases (ERK), p38, and c-Jun N-terminal kinase (JNK) pathways (or mitochondrial apoptosis pathway) and the caspase-3/ROCK1/MLC pathway; anti-inflammatory and immune stimulatory-related activities that involve apoptosis or necrosis via MAPK pathways (the JNK1/2 + ERK1/2 and PPARγ/HO-1 pathways), endoplasmic reticulum stress (ERS), high mobility group protein1 (HMGB1)-induced TLR2/4/9 and receptor for advanced glycation end products (RAGE) pathways, and the activation of NF-κB; neurological cell cycle, proliferation, differentiation, and regeneration via the MAPK pathways (JNK1/2 + ERK1/2, PI3K-Akt/mTOR, PKB/Akt and HIF-1α/VEGF pathways); and energy metabolism and the regulation of cellular ATP levels, the blood-brain barrier and other effects via N-methyl-D-aspartic acid (NMDA) receptors, ERS, and AMP/AMPK-GLUT pathways. Collectively, these mechanisms result in significant neuroprotective effects against cerebral ischemic injury. These findings will be valuable in that they should further promote the development of candidate drugs and provide more information to support the application of previous findings in stroke clinical trials.

Ischemic stroke and ischemia/reperfusion (I/R) injury induced by thrombolytic therapy are conditions with high mortality and serious long-term physical and cognitive disabilities. They have a major impact on global public health. These disorders are associated with multiple insults to the cerebral microcirculation, including reactive oxygen species (ROS) overproduction, leukocyte adhesion and infiltration, brain blood barrier (BBB) disruption, and capillary hypoperfusion, ultimately resulting in tissue edema, hemorrhage, brain injury and delayed neuron damage. Traditional Chinese medicine (TCM) has been used in China, Korea, Japan and other Asian countries for treatment of a wide range of diseases. In China, the usage of compound TCM preparation to treat cerebrovascular diseases dates back to the Han Dynasty. Even thousands of years earlier, the medical formulary recorded many classical prescriptions for treating cerebral I/R-related diseases. This review summarizes current information and underlying mechanisms regarding the ameliorating effects of compound TCM preparation, Chinese materia medica, and active components on I/R-induced cerebral microcirculatory disturbances, brain injury and neuron damage.

Multi-omics is a cutting-edge approach that combines data from different biomolecular levels, such as DNA, RNA, proteins, metabolites, and epigenetic marks, to obtain a holistic view of how living systems work and interact. Multi-omics has been used for various purposes in biomedical research, such as identifying new diseases, discovering new drugs, personalizing treatments, and optimizing therapies. This review summarizes the latest progress and challenges of multi-omics for designing new treatments for human diseases, focusing on how to integrate and analyze multiple proteome data and examples of how to use multi-proteomics data to identify new drug targets. We also discussed the future directions and opportunities of multi-omics for developing innovative and effective therapies by deciphering proteome complexity.
