Effects of Different Animal Manures on Aerobic Fermentation Process in Tobacco Stalk Composting

Abstract

To investigate the regulatory effects of different livestock manures on aerobic fermentation of tobacco stalks, composting experiments were conducted with tobacco stalks mixing separately with swine manure (T1), sheep manure (T2), cattle manure (T3), and chicken manure (T4) at an initial C/N ratio of 25:1. Key parameters including pile temperature, pH, electrical conductivity (EC), total organic carbon (TOC), total nitrogen (TN), carbon-to-nitrogen ratio (C/N), total phosphorus (TP), total potassium (TK), and germination index (GI) were dynamically monitored to evaluate manure-type effects on the maturation process. The results demonstrated that all treatments met thermophilic-phase sanitation standards, with post-fermentation (63 d) pH stabilizing between 8.00-8.50. Treatments T1 and T2 exhibited significantly lower EC but higher TOC than T3 and T4. Nutrient analysis showed that at the end of composting, the TN content in treatment T2 was significantly higher than that in T1, T3, and T4. The C/N ratios of all treatments eventually fell below 15. TP content peaked in T4 and reached minimum in T2, with TK accumulation following T4>T3>T1>T2. Maturity assessment showed significantly higher GI values in T1 and T2 versus T4, indicating superior compost maturity and reduced phytotoxicity. In summary, swine manure (T1) and sheep manure (T2) effectively enhanced organic matter conversion efficiency, optimized nutrient balance, and accelerated maturation, establishing them as optimal amendments for tobacco stalk valorization. This study provides critical theoretical and technical parameters for composting optimization.

Tobacco is widely planted across the world especially in China, which means that a large amount of tobacco waste needs to be treated. This study investigated the biogas fermentation of tobacco stalks co-digested with different biomass feedstocks and the inactivation of Tobacco mosaic virus (TMV), Cucumber mosaic virus (CMV) by anaerobic digestion. Results showed that the maximum methane yield of tobacco stalks at 35 °C was 0.163 m(3) CH4 ⋅ kg VS(-1), which was from the co-digestion of tobacco stalks, wheat stalks and pig manure. The largest VS removal rate of tobacco stalks was 59.10%. Proven by indicator paper stripe, half-leaf lesion and RT-PCR, CMV could be inactivated by mesophilic and thermophilic anaerobic digestion, whereas TMV could be only inactivated by thermophilic anaerobic digestion over 20 days. These results suggested that using tobacco stalks as feedstock for anaerobic digestion and applying the digested residue and slurry to Solanaceae crop land are feasible.Copyright © 2015. 为扩展烟草秸秆的利用途径,探究烟草秸秆绿原酸对烟草疫霉的抑菌效果,采用单因素试验和响应面分析法优化烟草秸秆绿原酸微波辅助提取工艺,并通过离体培养探讨烟草秸秆绿原酸对烟草疫霉菌的抑制作用。单因素及响应面结果表明,烟草秸秆绿原酸最佳提取工艺为料液比1∶15(g·mL<sup>-1</sup>),提取温度50.03 ℃,乙醇体积分数50%,此时粗提物中绿原酸含量为3.48 mg·g<sup>-1</sup>;离体条件下,烟草秸秆绿原酸的添加均抑制了疫霉菌的生长,其中175 mg·L<sup>-1</sup>烟草秸秆绿原酸对菌落直径的抑制效果最为显著,抑菌率高达32%。此外,碘化丙啶(PI)染色结果表明,175 mg·L<sup>-1</sup>的烟草秸秆绿原酸可通过破坏烟草疫霉菌细胞膜的完整性抑制菌丝生长。研究结果为烟草秸秆有效利用和新型天然植物源杀菌剂开发奠定了基础。

The seed germination index (GI) is a widely used indicator of compost maturity and is a required index in many national standards. However, seeds of different species vary markedly in sensitivity to the biological toxicity of compost. Therefore, suitable seed selection is essential for evaluation of compost maturity with the GI. This study systematically investigated the germination percentage and root length for seeds of 17 species incubated in deionized water for 48 h at 25 °C in the dark. Based on the germination percentage, seeds of eight species (white radish, cucumber, fruit radish, edible rape, round radish, hybrid cucumber, cress, and Chinese cabbage) were selected for determination of the GI of chicken manure composted with differing proportions of tobacco powder and mushroom substrate. The GI of hybrid cucumber seeds showed a significant positive correlation with temperature and pH, and a significant negative correlation with E/E ratio and ammonium-nitrogen content. The change in GI of hybrid cucumber seeds during composting and the GI value at the completion of composting were consistent with other maturity indicators. Among the tested seeds, the biological toxicity of the compost was best characterized by hybrid cucumber seeds, which thus represented a sensitive and reliable seed suitable for evaluation of compost maturity with the GI.Copyright © 2021 Elsevier Ltd. The present study determined the dynamic changes of bacterial community structures and its metabolism functions in rice straw composting process with different types of biochar addition. Results showed that both wheat and maize straw biochar addition greatly increased the temperatures and germination index. Biochar addition influenced microbial community structure and metabolic characteristics of the compost. Firmicutes, Actinobacteria and Proteobacteria were the dominant phyla throughout the process, and both biochar addition significantly increased the relative abundance of Actinobacteria phylum, especially genus Saccharomonospora. The abundance of bacterial genes related to amino acid metabolism, carbohydrate metabolism and energy metabolism were also increased with biochar addition. Moreover, total nitrogen was strongly affected by biochar addition and had the greatest influence on the bacterial community structure. These results indicated that biochar addition improved the maturity and fertility of the compost product as well as significantly regulated the microbial community structure and functions during composting process.Copyright © 2018 Elsevier Ltd. In this work, penicillin fermentation dregs (PFDs) and sewage sludge (SWS) were co-composted to analyze the possibility of recycling nutrients in PFDs. The temperature was maintained above 55°C for more than 3 days, and the final electrical conductivity (EC), pH and C/N all met the national standards in maturity. A nearly 100% removal of the residual penicillin was achieved, and the seed germination index (GI) increased from 0.02% to 83.54±3.1% by the end of the composting process. However, monitoring the quantity of antibiotic resistance genes (ARGs) showed that the logarithm of the number of copies of blaTEM increased from 4.17±0.19 at the initial phase to 8.92±0.27 by the end of the composting process, which means that there is a high risk for land use when using PFD compost products.Copyright © 2016 Elsevier Ltd. Compost is commonly used for the growth of plants and the remediation of environmental pollution. It is important to evaluate the quality of compost and seed germination test is a powerful tool to examine the toxicity of compost, which is the most important aspect of the quality. Now the test is widely adopted, but the main problem is that the test results vary with different methods and seed species, which limits the development and application of it. The standardization of methods and the modelization of seeds can contribute to solving the problem. Additionally, according to the probabilistic theory of seed germination, the error caused by the analysis and judgment methods of the test results can be reduced.
