Effects of Different Selenium Application Levels on Antioxidant and Detoxification of Rice

Abstract

This study aimed to investigate the effects of varying selenium (Se) concentrations on the antioxidant and detoxification responses in rice, and to provide a basis for subsequent research on Se-enriched rice. Using the rice cultivar ‘Meixiangzhan 2’ as the experimental material, a hydroponic experiment was conducted. Four Se concentration treatments (0, 0.01, 0.025, and 0.05 mg/L) were established by adding sodium selenite. Plant height, photosynthetic pigment content, selenium and nutrient (N, P, K) contents were determined, and changes in antioxidant enzyme (SOD, POD, CAT) activities, detoxification enzyme (GST, CarE) activities, and the expression level of rice selenium-binding protein 1 (OsSBP1) were analyzed. The results showed that appropriate Se application promoted rice growth, while excessive Se inhibited growth and caused dwarfing. The plant height was reduced by 21.8%-32.9% when increased Se concentration from 0.01 to 0.05 mg/L. Low Se concentrations had no effect on leaf photosynthetic pigment content, but the content decreased significantly at 0.05 mg/L. With increasing Se concentration, the Se content in rice roots, stems, and leaves continuously increased, reaching 236-, 163-, and 64-fold that of the control at 0.05 mg/L, respectively. However, the absorption and accumulation of nitrogen (N), phosphorus (P), and potassium (K) in rice were not inhibited. Malondialdehyde (MDA) content in roots stems and leaves decreased significantly with increasing Se levels. The lowest MDA values were observed at 0.05 mg/L (roots), 0.025 mg/L (stems), and 0.01 mg/L (leaves), with reductions of 79.5%, 65.7%, and 53.7%, respectively. Within the Se concentration range of 0.01-0.05 mg/L, the activities of superoxide dismutase (SOD), glutathione S-transferase (GST), and carboxylesterase (CarE) in roots, stems and leaves increased significantly. In contrast, the activities of peroxidase (POD) and catalase (CAT) in roots and stems initially increased and then decreased. The relative expression of OsSBP1 also showed a similar trend, decreasing to 0.5- and 0.3-fold the control level in stems and leaves at 0.05 mg/L, respectively. In summary, appropriate Se application increased the Se content, reduced MDA content, enhanced the activities of antioxidant and detoxification enzymes, and upregulated OsSBP1 expression in rice, thereby promoting antioxidant capacity and alleviating Se toxicity symptoms. However, excessive Se application inhibited antioxidant activity, exerted toxic effects, reduced photosynthetic pigment content, and suppressed the growth and development of rice. This study provides a theoretical reference for the rational application of Se fertilizer in rice cultivation.

Selenium (Se) is an important element that was discovered in 1817, and since the 1960s, it has been regarded as an essential micronutrient for both animals and humans, playing among other functions a relevant role in the antioxidant system of mammals. Inadequate blood Se levels in the human body is a well-known concern in many parts of the world. This malnutrition problem is often due to Se-poor diet, probably as a result of the low Se availability in soils where crops are growing. Nowadays, it is known that not only the total content, but also the inorganic and organic forms of Se contained in foods are important for human nutrition. However, paradoxically some regions of the world present high Se levels in soils causing several functional disorders and diseases in people who live in seleniferous areas. As essential micronutrient, Se should be supplied in controlled amounts to avoid harmful effects. Therefore, taking into account the importance of the soils as a way to ensure the adequate Se supply for the population, this review has dealt with Se behavior in soil environment (e.g., Se forms and adsorption and desorption reactions) and its relevance for the human health. As a final message, the further understanding of Se behavior in soils to predict its availability for crops in different systems will be a significant approach in future studies to establish forms and safe doses of Se to be added in fertilizers. Agronomic and genetic biofortification of staple crops with Se has been suggested as an important tool to improve Se intake by population in Se-poor sites.

Selenium (Se) can alleviate the toxicity of cadmium (Cd), but little is known about its mechanism in Cd uptake and translocation in plants. We investigated the effects of exogenous selenite, selenate, and selenomethionine (SeMet) on Cd uptake and translocation within rice (Oryza sativa L., Zhunliangyou 608) seedlings, and the concentration-dependent uptake kinetics of Cd into rice roots (with or without Se) were determined. The effect of the endogenous Se pool on Cd uptake was also investigated. Results of uptake kinetics showed that selenite slightly promoted Cd influx during 1h of exposure, compared with no selenite addition; Vmax of Cd uptake increased by 13.8% in 10μM selenite treatment; while the presence of selenate had no effect on the influx of Cd. When exposed to Cd (5μM) over 20h (with selenite) or 30h (with selenate or SeMet), Se addition (5μM) decreased Cd uptake and root-to-shoot translocation; after 30h selenite, selenate, or SeMet addition decreased Cd uptake by roots by 28.6%, 17.7% or 12.1%, respectively. Besides, as the selenite levels in the treatment solutions (1μMCd) increased (0, 0.1, 1, and 5μM, Se), Cd uptake and translocation were both significantly reduced, while the inhibitive effect was more significant at lower levels of selenate. Pretreatment of selenite or selenate (5μM) also decreased Cd uptake by 24.9% or 15.7%, and reduced the root-to-shoot transfer factor by 41.4% or 36.2% after 144h of subjection to Cd (5μM), respectively. The presence of selenite decreased Cd content more effectively than did selenate. Our results demonstrated that Se can effectively reduce the Cd translocation from roots to shoots in rice seedlings. 为研究两种不同外源硒施用对‘富源4号’水稻抗氧化酶活性、产量及其体内硒含量的影响,明确最佳的施硒方式,为富硒农产品发展提供理论指导。在宁夏低硒区(永宁县),以‘富源4号’水稻为试验材料,利用富硒微肥胶囊作土壤基施,螯合硒叶面肥结合在水稻关键生育时期作叶面喷施,采用数理统计等方法,比较分析不同外源施硒对水稻抗氧化酶活性、产量及其体内硒含量的变化规律。在水稻不同生长时期,水稻叶片MDA、POD活性分别比对照提高41.17%和35.98%,且螯合硒叶面肥处理与其他处理相比均有显著差异;叶片SOD活性呈现先增加后降低的趋势,各处理与对照相比差异显著且T3处理提高91.71%;不同施硒处理均提高了茎秆、叶片和大米全硒含量,大米有机硒含量约占到大米全硒含量的62.11%~80.96%,方差分析后与对照差异显著,且喷施效果好于基施。低硒区采用外源硒补给方式进行植物体内硒的富集有一定的效果,其中叶面喷施硒肥的效果要好于富硒微肥胶囊的结果,且缺硒区外源硒处理在一定程度上可提高水稻产量,同时能够增加提高水稻叶片及稻米中的全硒含量。

Selenium (Se) is an essential trace element for humans and animals, although controversial for different plant species. There exists a narrow line between essential, beneficial and toxic levels of Se to living organisms which greatly varies with Se speciation, as well as the type of living organisms. Therefore, it is crucial to monitor its solid- and solution-phase speciation, exposure levels and pathways to living organisms. Consumption of Se-laced food (cereals, vegetables, legumes and pulses) is the prime source of Se exposure to humans. Thus, it is imperative to assess the biogeochemical behavior of Se in soil-plant system with respect to applied levels and speciation, which ultimately affect Se status in humans. Based on available relevant literature, this review traces a plausible link among (i) Se levels, sources, speciation, bioavailability, and effect of soil chemical properties on selenium bioavailability/speciation in soil; (ii) role of different protein transporters in soil-root-shoot transfer of Se; and (iii) speciation, metabolism, phytotoxicity and detoxification of Se inside plants. The toxic and beneficial effects of Se to plants have been discussed with respect to speciation and toxic/deficient concentration of Se. We highlight the significance of various enzymatic (catalase, peroxidase, superoxide dismutase, ascorbate peroxidase, glutathione peroxidase) and non-enzymatic (phytochelatins and glutathione) antioxidants which help combat Se-induced overproduction of reactive oxygen species (ROS). The review also delineates Se accumulation in edible plant parts from soils containing low or high Se levels; elucidates associated health disorders or risks due to the consumption of Se-deficient or Se-rich foods; discusses the potential role of Se in different human disorders/diseases.Copyright © 2017 Elsevier Ltd. The present study investigated the phenotypical, physiological and biochemical changes of rice plants exposed to high selenium (Se) concentrations to gain an insight into Se-induced phytotoxicity. Results showed that exposure of rice plants to excessive Se resulted in growth retardation and biomass reduction in connection with the decreased levels of chlorophyll, carotenoids and soluble proteins. The reduced water status and an associated increase in sugar and proline levels indicated Se-induced osmotic stress in rice plants. Measurements of Se contents in roots, leaf sheaths and leaves revealed that Se was highly accumulated in leaves followed by leaf sheaths and roots. Se also potentiated its toxicity by impairing oxidative metabolism, as evidenced by enhanced accumulation of hydrogen peroxide, superoxide and lipid peroxidation product. Se toxicity also displayed a desynchronized antioxidant system by elevating the level of glutathione and the activities of superoxide dismutase, glutathione-S-transferase and glutathione peroxidase, whereas decreasing the level of ascorbic acid and the activities of catalase, glutathione reductase and dehydroascorbate reductase. Furthermore, Se triggered methylglyoxal toxicity by inhibiting the activities of glyoxalases I and II, particularly at higher concentrations of Se. Collectively, our results suggest that excessive Se caused phytotoxic effects on rice plants by inducing chlorosis, reducing sugar, protein and antioxidant contents, and exacerbating oxidative stress and methylglyoxal toxicity. Accumulation levels of Se, proline and glutathione could be considered as efficient biomarkers to indicate degrees of Se-induced phytotoxicity in rice, and perhaps in other crops.Copyright © 2017 Elsevier Ltd. Selenium is an essential trace element in the human body. However, its intake is generally low. Therefore, the production and utilisation of selenium-enriched foods is currently a research hotspot. In this study, the effects of low (0.2 mg·kg−1), medium (1.0 mg·kg−1), and high (5.0 mg·kg−1) concentrations of selenium on the physiological and biochemical characteristics of rice were investigated to develop selenium-enriched rice. High concentrations of selenium have been found to inhibit the growth, physiology, and biochemistry of rice, while low concentrations of selenium promote its growth. The height of mature rice plants exposed to high concentrations of selenium was reduced by 7.20% compared with the height of control rice. Selenium decreased the proline content of rice during the growth period except in mature rice treated with medium and high concentrations of selenium. Excluding high concentrations, selenium treatment increased the soluble sugar content of rice from the tillering to the mature stages. The peroxidase activity of rice at the heading stage treated with medium levels of selenium was significantly higher than that of the control rice, while the superoxide dismutase activity of rice exposed to selenium was significantly enhanced at the mature stage. The malondialdehyde levels of mature rice treated with medium and high levels of selenium were significantly lower than those of the control rice. The selenium content of each plant part was significantly correlated with the soil selenium level. An increase in the soil selenium level facilitated the production of selenium-enriched rice.

<p>Greenhouse experiment was conducted to investigate selenium accumulation and its antioxidant response in two rice varieties (PR116 and Pusa Basmati 1121) grown on normal and seleniferous soils. The plant growth was reduced at early developmental stages and flowering was delayed by a period of 10 d on seleniferous soil. Selenium accumulation increased by 3&ndash;20 and 13&ndash;14 folds in leaves, 18 and 3 folds in grains from Pusa Basmati 1121 and PR116 varieties, respectively. Selenium accumulation in leaves from rice plants grown on seleniferous soil resulted in significant increase in chlorophyll content, hydrogen peroxide, proline, free amino acids, total phenol and tannin contents. Lipid peroxidation levels and peroxidase activities in leaves increased whereas catalase activity showed a reverse trend. It is concluded that selenium accumulation decreased dry matter content in rice during crop development but these plants were able to combat selenium toxicity by inducing alterations in their defense system.</p>

Selenium-binding proteins (SBPs) represent a ubiquitous and conserved protein family with yet unclear biochemical and molecular functions. The importance of the human homolog has been extensively studied as it is implicated in many cancer types and other diseases. On the other hand, little is known regarding plant homologs. In plants, there is evidence that SBP participates in developmental procedures, oxidative stress responses, selenium and cadmium binding, and pathogenic tolerance. Moreover, recent studies have revealed that SBP is a methanethiol oxidase (MTO) catalyzing the conversion of methanethiol into formaldehyde, H2S, and H2O2. The two later products emerge as key signal molecules, playing pivotal roles in physiological processes and environmental stress responses. In this review, we highlight the available information regarding plants in order to introduce and emphasize the importance of SBP1 and its role in plant growth, development, and abiotic/biotic stress.

In Arabidopsis (Arabidopsis thaliana), the putative selenium-binding protein (SBP) gene family is composed of three members (SBP1-SBP3). Reverse transcription-polymerase chain reaction analyses showed that SBP1 expression was ubiquitous. SBP2 was expressed at a lower level in flowers and roots, whereas SBP3 transcripts were only detected in young seedling tissues. In cadmium (Cd)-treated seedlings, SBP1 level of expression was rapidly increased in roots. In shoots, SBP1 transcripts accumulated later and for higher Cd doses. SBP2 and SBP3 expression showed delayed or no responsiveness to Cd. In addition, luciferase (LUC) activity recorded on Arabidopsis lines expressing the LUC gene under the control of the SBP1 promoter further showed dynamic regulation of SBP1 expression during development and in response to Cd stress. Western-blot analysis using polyclonal antibodies raised against SBP1 showed that SBP1 protein accumulated in Cd-exposed tissues in correlation with SBP1 transcript amount. The sbp1 null mutant displayed no visible phenotype under normal and stress conditions that was explained by the up-regulation of SBP2 expression. SBP1 overexpression enhanced Cd accumulation in roots and reduced sensitivity to Cd in wild type and, more significantly, in Cd-hypersensitive cad mutants that lack phytochelatins. Similarly, in Saccharomyces cerevisiae, SBP1 expression led to increased Cd tolerance of the Cd-hypersensitive ycf1 mutant. In vitro experiments showed that SBP1 has the ability to bind Cd. These data highlight the importance of maintaining the adequate SBP protein level under healthy and stress conditions and suggest that, during Cd stress, SBP1 accumulation efficiently helps to detoxify Cd potentially through direct binding.

The function of selenium-binding protein 1 (SBP1), present in almost all organisms, has not yet been established. In mammals, SBP1 is known to bind the essential element selenium but the binding site has not been identified. In addition, the SBP family has numerous potential metal-binding sites that may play a role in detoxification pathways in plants. In Arabidopsis thaliana, AtSBP1 over-expression increases tolerance to two toxic compounds for plants, selenium and cadmium, often found as soil pollutants. For a better understanding of AtSBP1 function in detoxification mechanisms, we investigated the chelating properties of the protein toward different ligands with a focus on selenium using biochemical and biophysical techniques. Thermal shift assays together with inductively coupled plasma mass spectrometry revealed that AtSBP1 binds selenium after incubation with selenite (SeO3(2-)) with a ligand to protein molar ratio of 1:1. Isothermal titration calorimetry confirmed the 1:1 stoichiometry and revealed an unexpectedly large value of binding enthalpy suggesting a covalent bond between selenium and AtSBP1. Titration of reduced Cys residues and comparative mass spectrometry on AtSBP1 and the purified selenium-AtSBP1 complex identified Cys(21) and Cys(22) as being responsible for the binding of one selenium. These results were validated by site-directed mutagenesis. Selenium K-edge x-ray absorption near edge spectroscopy performed on the selenium-AtSBP1 complex demonstrated that AtSBP1 reduced SeO3(2-) to form a R-S-Se(II)-S-R-type complex. The capacity of AtSBP1 to bind different metals and selenium is discussed with respect to the potential function of AtSBP1 in detoxification mechanisms and selenium metabolism.© 2014 by The American Society for Biochemistry and Molecular Biology, Inc.
