Isolation and Identification of Pathogen Causing Black Mold Disease on Ginger During Storage and Study on Its Biological Characteristics

Isolation and Identification of Pathogen Causing Black Mold Disease on Ginger During Storage and Study on Its Biological Characteristics
This study aimed to identify the pathogen causing black mold on stored ginger in Sichuan and clarify its key biological characteristics, so as to provide a theoretical basis for green control strategies. The pathogenic strain CDSJ1 was obtained by tissue isolation, and identified based on morphological characteristics, molecular biology (ITS sequence analysis), and verification using Koch’s postulates. The effects of temperature, carbon source, nitrogen source, and light conditions on mycelial growth and sporulation were investigated through single-factor experiments. The pathogen was identified as Aspergillus niger. The optimal growth temperature was 30 ℃; the most suitable carbon and nitrogen sources were maltose and tryptone, respectively; and the highest sporulation level was observed under continuous light conditions. This study systematically identifies A. niger as the pathogen of black mold on stored ginger in Sichuan and elucidate its key biological characteristics, providing a scientific basis for the development of green postharvest disease management strategies.

The aim of this study was to investigate differences in the dynamics of ochratoxin A (OTA) production by various Aspergillus isolates under different cultivation conditions. Nine strains representing A. westerdijkiae, A. ochraceus, A. sulphureus, A. carbonarius, and A. albertensis were tested on malt extract agar (MEA), Czapek yeast extract agar (CYA), potato dextrose agar (PDA), and yeast extract sucrose agar (YES). Cultivations were performed at 18 °C, 22 °C, 25 °C, and 30 °C, and OTA production was monitored on the 6th, 10th, 14th, 21st, and 30th days using HPLC analysis. OTA yields strongly depended on the producing strain, with significant variability even among isolates of the same species. The most productive strain was A. ochraceus from cereals with a maximum concentration of 848 µg g−1 OTA, followed by two isolates of A. westerdijkiae from grapes of Slovak origin (591 and 479 µg g−1), and A. sulphureus from soil (546 µg g−1). In contrast, A. carbonarius strains showed the weakest OTA production. Across media, YES supported the highest toxin levels, whereas the most favourable cultivation temperatures were 18 °C and 25 °C. Each strain reached its production maximum at different time points, highlighting the strain-specific nature of OTA biosynthesis.

从草地贪夜蛾成虫尸体中分离获得一株杀虫真菌。经形态和分子鉴定,确定该虫生真菌为莱氏绿僵菌,并命名为NrSfadult202104。生物学特性的研究结果显示,该菌株在SMAY+0.5%草地贪夜蛾蛹粉培养基上生长速率和分生孢子产量最佳;在28 ℃时菌落生长速率最快,20 ℃时产孢最为适宜;在全光照条件下该菌株产孢量最高。对草地贪夜蛾成虫和3龄幼虫的致病力测定结果显示,该菌株对草地贪夜蛾雌、雄成虫以及3龄幼虫都具有一定的毒力效果。当孢子浓度为1.0×10<sup>8</sup>孢子/mL时,其感染雌、雄成虫校正死亡率分别为61.2%和58.1%;菌株接种3龄幼虫后,随孢子悬浮液浓度的升高,幼虫的感病死亡率也随之增加,当浓度达到1.0×10<sup>8</sup>孢子/mL时,3龄幼虫的致死中时(LT<sub>50</sub>)为5.30 d,接种8 d后3龄幼虫的校正死亡率为66.12%。综合以上结果,莱氏绿僵菌NrSfadult202104菌株对草地贪夜蛾具有潜在的生防作用,值得深入研究。
