Effects of 4,8-DHT on Weeds Inhibition and Physiological-Biochemical Parameters
To explore the herbicidal mechanism of 4,8-dihydroxy-1-tetrahydronaphthone (4,8-DHT), this study carried out a weed seed germination experiment to assess the inhibitory effects of four concentrations of 4,8-DHT (0.5, 1.0, 2.5, and 5.0 mmol/L) on nine weed species (Lindernia procumbens, Eleusine indica, Salvia japonica, Taraxacum mongolicum, Avena fatua, Poa annua, Digitaria sanguinalis, Cynodon dactylon, Echinochloa crusgalli). In accordance with industry standards, representative weeds (L. procumbens and A. fatua) that meet germination rate criteria and sensitivity to 4,8-DHT were selected. Physiological-biochemical parameters analyses and transcriptomic profiling were employed to further reveal their potential mechanisms of action. The results demonstrated that high concentrations of 4,8-DHT (>2.5 mmol/L) significantly inhibited the seed germination of all nine tested weed species, and the inhibitory effect was concentration-dependent. At 5.0 mmol/L, 4,8-DHT exhibited inhibition rate of 89.9% and 86.3% for L. procumbens and A. fatua, respectively. The physiological-biochemical analyses revealed that as the concentration of 4,8-DHT increased, the activities of peroxidase (POD) and catalase (CAT) in the weeds exhibited a pattern of initially increasing and subsequently decreasing. Meanwhile, the levels of malondialdehyde (MDA) and proline (Pro) continuously increased, indicating that 4,8-DHT might impede the physiological activities of the weeds by interfering with the antioxidant defense system and the metabolism of osmotic regulatory substances. Transcriptomic analysis further disclosed that after treating L. procumbens with 4,8-DHT, a substantial number of differentially expressed genes were generated. Among these, the treatment group at 5 mmol/L (3295 genes) was significantly larger than that at 0.5 mmol/L (1858 genes), and these genes were mainly enriched in pathways including glutathione metabolism, glyoxylate and dicarboxylate metabolism, carbon fixation pathways, and plant-pathogen interactions. The research findings uncovered the potential mechanism by which 4,8-DHT inhibits the growth of weeds at the physiological-biochemical levels, offering scientific guidance for the development and practical application of 4,8-DHT as a novel plant-based herbicide.

Modern herbicides make major contributions to global food production by easily removing weeds and substituting for destructive soil cultivation. However, persistent herbicide selection of huge weed numbers across vast areas can result in the rapid evolution of herbicide resistance. Herbicides target specific enzymes, and mutations are selected that confer resistance-endowing amino acid substitutions, decreasing herbicide binding. Where herbicides bind within an enzyme catalytic site very few mutations give resistance while conserving enzyme functionality. Where herbicides bind away from a catalytic site many resistance-endowing mutations may evolve. Increasingly, resistance evolves due to mechanisms limiting herbicide reaching target sites. Especially threatening are herbicide-degrading cytochrome P450 enzymes able to detoxify existing, new, and even herbicides yet to be discovered. Global weed species are accumulating resistance mechanisms, displaying multiple resistance across many herbicides and posing a great challenge to herbicide sustainability in world agriculture. Fascinating genetic issues associated with resistance evolution remain to be investigated, especially the possibility of herbicide stress unleashing epigenetic gene expression. Understanding resistance and building sustainable solutions to herbicide resistance evolution are necessary and worthy challenges.

As a candidate for bioherbicide, 4,8-dihydroxy-1-tetralone (4,8-DHT) was isolated from Caryospora callicarpa epicarp and its two enantiomers, S-(+)-isosclerone and R-(−)-regiolone, were separated by chiral high-performance liquid chromatography (HPLC) on a Chiralcel OD column with chiral stationary phase (CSP)-coated cellulose-tris(3,5-dimethylphenylcarbamate). Then, the phytotoxicity of 4,8-DHT and its enantiomers toward the seeds germination and seedling growth of the five tested plant species, including lettuce (Latuca sativa), radish (Raphanus sativus), cucumber (Cucumis sativus), onion (Allium cepa), and wheat (Triticum aestivum), were investigated and the results indicated a hormesis at low concentration of 4,8-DHT and its enantiomers, but a retardant effect at high concentration. Between the two enantiomers of 4,8-DHT, the S-(+)-isosclerone was more toxic to seeds germination and seedling growth of the five tested plant species than the R-(−)-regiolone, and also the phytotoxicity of S-(+)-isosclerone varied with different plants. For example, S-(+)-isosclerone was the most active to seedling growth of lettuce, indicating that S-(+)-isosclerone had specific effects on different organisms. Thus, all of the chirality and concentration of 4,8-DHT, as well as the affected plant species, need to be taken into consideration in the development and utilization of 4,8-DHT.

The alignment of sequencing reads against a protein reference database is a major computational bottleneck in metagenomics and data-intensive evolutionary projects. Although recent tools offer improved performance over the gold standard BLASTX, they exhibit only a modest speedup or low sensitivity. We introduce DIAMOND, an open-source algorithm based on double indexing that is 20,000 times faster than BLASTX on short reads and has a similar degree of sensitivity.

Phytotoxic substances in plants that may serve as alternative natural herbicides for controlling weeds are required for sustainable agriculture. We explored the phytotoxic activities of aqueous methanol extracts of Senna garrettiana (Craib) Irwin & Barneby leaves and the active substances they contain. The results revealed that the S. garrettiana leaf extracts had significant phytotoxic effects on three dicotyledons (Lepidium sativum L., Lactuca sativa L., and Medicago sativa L.) and two monocotyledons (Phleum pratense L. and Lolium multiflorum Lam.). An bioassay-guided isolation process yielded three active substances; caffeic acid, methyl caffeate, and (S)-6-hydroxymellein inhibited the seed germination, seedling growth, and biomass accumulation of L. sativum in a concentration-dependent manner. Based on the concentration required for 50% growth inhibition (IC50), (S)-6-hydroxymellein had the highest inhibitory effects on L. sativum in all test parameters, followed by methyl caffeate and caffeic acid. The L. sativum roots were the most susceptible to (S)-6-hydroxymellein (IC50 = 383 µM) and caffeic acid (IC50 = 2627 µM), whereas methyl caffeate (IC50 = 1361 µM) had the greatest effect on the L. sativum shoots. Thus, three isolated compounds may explain the phytotoxic effects of the S. garrettiana extracts. Consequently, caffeic acid, methyl caffeate, and (S)-6-hydroxymellein could be potential candidates for the future production of bioherbicides.
